Thyrotropin cross - links to the thyrotropin receptor through both the a and subunits Paul

چکیده

We have recently shown that the /3 subunit of thyrotropin (TSH) can be cross-linked to the TSH receptor [Buckland, Strickland, Pierce & Rees Smith (1985) Endocrinology (Baltimore) 116, 2122-2124; Buckland, Strickland & Rees Smith (1985) Biochem. Soc. Trans. 13, 942-943]. We failed, however, to cross-link the a subunit to the receptor; leaving the role of this subunit in the TSH-TSH-receptor interaction uncertain. We now report the successful cross-linking of the TSH a subunit to the receptor by the use of two different cross-linking reagents. Our studies suggest therefore that both subunits of TSH form part of the hormone's receptor-binding site.

The measurement technologies of thyrotropin receptor antibodies from the past to the present

Thyroid Stimulating Hormone Receptor (TSH-R) autoantibodies are the main cause of Graveschr('39') disease and its external thyroid manifestations such as ophtalmopathy and dermatopathy. These antibodies are functionally different and are commonly called TSH receptor antibodies (TRAbs). In fact, TRAbs are a set of autoantibodies including TSHR-stimulating antibodies (TSAbs), TSHR- blocking antib...

The Therapeutic Effect of Thyrotropin Releasing Hormone in Amyotrophic Sclerosis

Although the issue has not yet been fully resolved, there is reason to believe that TRI-I may have a trophic effect on the normal motoneuron and may transien tly improve the neurologic deficit in ALS. Since ALS is not a simple TRI-I deficiency disease, this improvement probably occurs via a secondary mecha -nism-possibly another neurotransmitter. It is uncertain at this time whether this effec...

THYROXINE AND THYROTROPIN LEVELS IN CORD BLOOD SERA FROM SOUTH OF IRAN

Birth is associated with significant changes in the hypothalamic-pituitarythyroid axis and these changes complicate the proper assessment of thyroid function in neonates. Normal values forT4 (thyroxine) and TSH (thyroid stimulating hormone) in cord blood are not well established. The present study was carried out on 4000 specimens of cord sera of both sexes in order to establish reliable re...

Affinity-labelling of the thyrotropin receptor

Thyrotropin (TSH) has been coupled to the photoactive heterobifunctional reagent N-hydroxysuccinimidyl 4-azidobenzoate (HSAB) and the properties of the product (HSAB-TSH) investigated. Preparations of HSAB-TSH containing two molecules of HSAB per molecule of TSH were used in most experiments and these preparations retained about 40% of the original receptor-binding activity of the TSH. HSAB-TSH...

The Gar , and Garll Proteins Couple the Thyrotropin - releasing Hormone Receptor to Phospholipase C in GH 3 Rat Pituitary Cells

Thyrotropin-releasing hormone stimulates the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP4 in GH3 cell membranes. The stimulation of the phosphoinositide phospholipase C (PI/PLC) activity can be blocked by incubation of GH3 membranes with polyclonal antibodies directed against a peptide derived from the C-terminal region of Gaq and G a l l . Antibodies directed against the C-termina...
